FKHR activity is tightly regulated by various post-translational modifications, including phosphorylation, acetylation, and ubiquitination. One of the key regulators of FKHR is the PI3K/AKT pathway. When activated, AKT phosphorylates FKHR, leading to its sequestration in the cytoplasm and preventing it from activating target genes in the nucleus. Conversely, when the PI3K/AKT pathway is inhibited, FKHR translocates to the nucleus and activates gene transcription.
