How is the Hedgehog Pathway Aberrantly Activated in Cancer?
There are several mechanisms through which the Hedgehog pathway can become aberrantly activated in cancer:
Mutations in PTCH1 or SMO that cause constitutive pathway activation. Overexpression of Hedgehog ligands, leading to autocrine or paracrine signaling. Loss of function mutations in negative regulators of the pathway.
