How is the Wnt Pathway Altered in Cancer?

Dysregulation of the Wnt pathway is a common feature in various cancers. Mutations in components of the pathway, such as APC, β-catenin, and AXIN, can lead to aberrant activation of Wnt signaling. This results in uncontrolled cell proliferation and survival, contributing to tumorigenesis. For instance, mutations in the APC gene are frequently observed in colorectal cancer, leading to the accumulation of β-catenin in the nucleus and activation of oncogenic target genes.
