Aberrant activation of the Hedgehog pathway is implicated in the development and progression of several types of cancer. Mutations in pathway components such as PTCH or SMO, or the overexpression of Hedgehog ligands, can lead to uncontrolled cell proliferation and survival. This has been observed in cancers like basal cell carcinoma (BCC), medulloblastoma, and some gastrointestinal cancers.
