STAT3 is activated through phosphorylation by upstream kinases such as Janus Kinases (JAKs) and Src family kinases. This phosphorylation event leads to STAT3 dimerization, nuclear translocation, and subsequent transcriptional activation of target genes. Aberrant activation can result from mutations, overexpression of upstream kinases, or increased levels of cytokines like IL-6.
