What types of p53 mutations are commonly seen in cancers?
The majority of the mutations in the TP53 gene are missense mutations, which result in the substitution of a single amino acid in the p53 protein. These mutations often occur in the DNA-binding domain, impairing the protein's ability to regulate target genes effectively. Some of the most common hotspot mutations include R175H, R273H, and R248Q. These hotspot mutations are recurrent across various types of cancers and are associated with particularly aggressive disease phenotypes.
