How Do Mutations in the TATA Box Contribute to Cancer?
Mutations in the TATA box can impair the binding of TBP, leading to aberrant transcription initiation. Such mutations may cause either overexpression or underexpression of oncogenes and tumor suppressor genes, respectively. For instance, a mutation that weakens TBP binding might reduce the transcription of a tumor suppressor gene, thereby promoting cancer development.
