Specificity: Many of the enzymes and pathways involved in acidification are also present in normal tissues, raising concerns about off-target effects. Delivery: Effective delivery of therapeutic agents to the tumor site remains a significant hurdle. Heterogeneity: Tumors are highly heterogeneous, and not all cancer cells within a tumor may exhibit the same degree of reliance on glycolysis or proton export mechanisms.
