One of the main challenges in targeting TNFR1 is its dual role in cancer. While it can induce apoptosis, it can also promote inflammation and a supportive tumor microenvironment. Another challenge is the ubiquitous expression of TNFR1, which could lead to systemic side effects if targeted therapeutically. Therefore, selective targeting strategies and a deeper understanding of TNFR1's context-dependent roles are essential.
